The present study aimed to study the efficiency of 1% Hydrochloric acid ethanol for decolorization in Hematoxylin and Eosin (H&E) stained rat tissues. Twelve tissues from rat were stained with H&E and stored at room temperature for 7 days. The triplicate slides stained tissues were decolorized with 1% Hydrochloric acid ethanol at 2, 4, 6, 8 and 10 minutes and photographed at 100 magnification in the similar positions. The pictures were analyzed with Fiji ImageJ program to determine the remaining of H&E and the average time for decolorization of each tissues were calculated. The results showed the efficiency of the reagents into 3 different groups. The reagent is most effective in skin, heart, brain and testis tissues, with an average time of 2 minutes. The reagent is moderate effective in liver, kidney, stomach and lung tissues, with an average time of 9.5 minutes. The reagent is less effective in small intestine, spleen, ovary, and pancreas tissues, with more than 10 minutes. In addition, the reagent is not caused tissues damaging or tissues detachment from slide. This reagent is actually valuable in file of histopathology study which can use the same slide to stain with other staining in order to add more information and provide more accuracy for interpretation in histopathological study.
